Abstract

Papillary thyroid carcinoma (PTC) and Hashimoto’s thyroiditis (HT) appear antagonistic but often occur simultaneously. This study integrates four GEO datasets to compare gene expression differences among PTC, HT, PTC_ HT, and control thyroid tissues (Control), revealing similarities and differences in gene expression. Results show significant differences in 202 genes among the three conditions, with consistent upregulation/downregulation trends, primarily involving inflammation and chemokines (e.g., CXCL8, CXCL9, CXCL10, IL8). Cells in PTC and HT are in a state of chronic inflammatory activation and are unresponsive to p53 and FAS-induced apoptotic signals. Analysis of differentially expressed genes between PTC and HT reveals that several extracellular matrix pathway genes (e.g., FN1, LAMB1, LAMC1) have opposite expression trends in the two diseases, and the FN1-DUSP6-ETV5-NFATC1/3 signaling axis may drive phenotypic heterogeneity by regulating T cell immune responses. These findings provide new insights into the molecular mechanisms and interrelationship of PTC and HT.
